TgAP2X-5 is a cell cycle-regulated ApiAP2 transcription factor in Toxoplasma gondii that acts within a cooperative transcriptional complex to drive expression of S/M-phase virulence genes (PMID:29788176). It was identified as a physical interaction partner of TgAP2XI-5 and functions to enable that factor's promoter occupancy: loss of AP2X-5 impairs AP2XI-5 binding across multiple rhoptry gene promoters genome-wide (PMID:29788176). Consistent with this regulatory role, inducible knockdown of AP2X-5 reduces transcript levels of numerous virulence factors, including rhoptry and microneme genes (PMID:29788176). This transcriptional deficit has a sharply compartmentalized functional consequence: AP2X-5 disruption only mildly affects host-cell invasion in vitro but renders parasites completely avirulent in mice, establishing it as an in vivo determinant of virulence (PMID:29788176).

  1. 2018 Medium

Establishing that AP2X-5 does not act alone but partners with another ApiAP2 factor defined it as a component of a cooperative transcription factor complex rather than an independent regulator.

Evidence Co-immunoprecipitation / interaction screen identifying AP2X-5 as a binding partner of AP2XI-5 in T. gondii

    PMID:29788176

    Open questions at the time
    • Single Co-IP without reciprocal validation or stoichiometry of the complex
    • Whether the interaction is direct or bridged by other factors is unresolved
    • DNA-binding specificity of AP2X-5 itself not defined
  2. 2018 Medium

Conditional knockdown coupled to transcriptomics answered whether AP2X-5 is functionally required for gene expression, showing it controls a defined virulence-gene program.

Evidence Inducible conditional knockdown plus RNA-seq in T. gondii

    PMID:29788176

    Open questions at the time
    • Does not distinguish direct AP2X-5 targets from downstream transcriptional effects
    • Full set of dysregulated loci and their promoter architecture not detailed here
  3. 2018 Medium

Genome-wide ChIP of AP2XI-5 in an AP2X-5-depleted background answered the mechanistic question of how AP2X-5 acts, revealing it is required for the partner factor's promoter occupancy at virulence loci.

Evidence Genome-wide ChIP of AP2XI-5 in the AP2X-5 knockdown strain

    PMID:29788176

    Open questions at the time
    • Whether AP2X-5 itself binds these promoters directly not shown
    • Mechanism by which AP2X-5 stabilizes AP2XI-5 occupancy is unknown
  4. 2018 Medium

Comparing in vitro invasion with in vivo mouse infection resolved the biological consequence of AP2X-5 loss, showing its requirement is specific to virulence rather than basic invasion.

Evidence Mouse infection virulence assay and in vitro invasion assay with AP2X-5 conditional knockdown parasites

    PMID:29788176

    Open questions at the time
    • Which specific dysregulated virulence gene(s) account for the avirulence not pinpointed
    • Effect across parasite strains/lineages not tested
  5. 2018 Low

Evidence Expression/cell cycle staging of AP2X-5 protein and mRNA

    PMID:29788176

    Open questions at the time
    • Correlative timing only, without functional dissection of the cell cycle dependence
    • Upstream regulators controlling AP2X-5 periodic expression unknown

Open questions

Synthesis pass · forward-looking unresolved questions
  • The intrinsic DNA-binding specificity of AP2X-5, the direct versus indirect nature of its target repertoire, and the structural basis of its cooperation with AP2XI-5 remain undefined.
  • No DNA-binding motif or direct genome-wide occupancy map for AP2X-5 itself
  • No structural model of the AP2X-5/AP2XI-5 complex
  • Requirement across parasite strains not established
