Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

TgAP2XI-5 is a cell cycle-regulated ApiAP2 transcription factor in Toxoplasma gondii that binds a defined cis-regulatory DNA element at more than 300 gene promoters and activates transcription of genes with peak expression in the S and M phases of the tachyzoite cell cycle, including virulence factors packaged in micronemes and rhoptries (PMID:24025328). Full binding at rhoptry gene promoters requires cooperation with a second ApiAP2 factor, TgAP2X-5: the two proteins co-immunoprecipitate, and knockdown of TgAP2X-5 abolishes TgAP2XI-5 promoter occupancy at multiple rhoptry genes (PMID:29788176). AP2XI-5 protein expression peaks in a defined temporal window during the tachyzoite cell cycle (S/M-subtranscriptome phase), consistent with its mRNA expression pattern, suggesting cell cycle-regulated protein delivery mediated by AP2 domain promoter recognition specificity (PMID:20865045). Beyond these promoter-binding and partner findings, no structural model or catalytic mechanism has been characterized in the available corpus.

Mechanistic history

Synthesis pass · year-by-year structured walk · 3 steps
  1. 2010 Low

Establishing when AP2XI-5 acts required knowing its expression timing; profiling showed protein and mRNA peak in a defined S/M cell cycle window, framing it as a candidate temporal regulator of that subtranscriptome.

Evidence cell cycle transcriptomic profiling and protein expression timing analysis in tachyzoites

    PMID:20865045

    Open questions at the time
    • Expression-based inference only, with no direct functional experiment on AP2XI-5
    • Does not identify target genes or DNA-binding specificity
  2. 2013 Medium

Whether AP2XI-5 is a bona fide transcriptional regulator with defined targets was open; ChIP plus cis-element identification showed it binds >300 promoters via a specific DNA motif and drives transcription of S/M-phase virulence genes, establishing it as a sequence-specific activator.

Evidence ChIP, promoter binding assays, cis-element identification, transcriptomic and reporter analysis in T. gondii

    PMID:24025328

    Open questions at the time
    • Single-lab study without genome-wide ChIP-seq resolution
    • Direct activation shown by reporter but native regulatory mechanism not dissected
  3. 2018 High

Whether AP2XI-5 acts alone at virulence promoters was unknown; reciprocal Co-IP and ChIP-seq showed it partners with TgAP2X-5, and TgAP2X-5 knockdown abolishes AP2XI-5 binding at multiple rhoptry gene promoters, establishing cooperative combinatorial control of virulence gene expression.

    Evidence reciprocal Co-IP, inducible knockdown, RNA-seq and genome-wide ChIP-seq in T. gondii

    PMID:29788176

    Open questions at the time
    • Mechanism of cooperativity (adjacent motifs vs. protein-protein tethering) not resolved
    • Does not address microneme promoters equivalently to rhoptry promoters

2013 TgAP2XI-5 binds to more than 300 gene promoters and actively controls transcription of genes with peak expression in S and M phases of the tachyzoite cell cycle, including virulence factors packaged in micronemes and rhoptries. The cis-regulatory element recognized by TgAP2XI-5 was identified, and it was demonstrated to actively drive gene transcription. 2018 TgAP2XI-5 interacts with TgAP2X-5 (identified by co-immunoprecipitation and proteomics). Disruption of TgAP2X-5 leads to defects in TgAP2XI-5 binding at multiple rhoptry gene promoters, demonstrating cooperative binding of these two ApiAP2 transcription factors is required for proper virulence gene expression.
