Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

SRS9 is a developmentally regulated, bradyzoite-specific member of the SAG1-related sequence (SRS) family of putative surface proteins in Toxoplasma gondii, upregulated during the tachyzoite-to-bradyzoite differentiation that underlies chronic infection (PMID:12455982). It is the dominant surface antigen recognized by the bradyzoite-specific P36 monoclonal antibody (PMID:17368655). Functionally, SRS9 contributes to the maintenance of parasite persistence in the brain and to the control of reactivation: deletion of SRS9 reduces brain cyst loads during chronic infection and accelerates intestinal reactivation in immunosuppressed hosts (PMID:17261600). Its strict stage restriction is functionally important as an immune-evasion mechanism—forced constitutive expression of SRS9 in tachyzoites elicits an SRS9-specific IFN-γ response that progressively eliminates persisting cysts (PMID:15944311). This tachyzoite-stage repression is enforced transcriptionally by the ApiAP2 factor AP2IV-4, whose deletion causes ectopic SRS9 expression in replicating tachyzoites (PMID:29718996), and the SRS9 promoter is itself a functional driver of bradyzoite-stage gene expression (PMID:31572690). Beyond these surface-antigen and stage-regulation roles, no biochemical activity or molecular interaction partner of the SRS9 protein has been characterized in the available corpus.

Mechanistic history

Synthesis pass · year-by-year structured walk · 6 steps
  1. 2002 Medium

Established that SRS9 exists as a stage-regulated gene, framing it as a candidate surface protein of the developmental switch rather than a constitutive antigen.

    • Family membership inferred from sequence similarity, not biochemical characterization
  2. 2005 High

Answered why SRS9 expression is stage-restricted, showing that confining it to bradyzoites shields the parasite from an SRS9-specific immune response that otherwise clears persisting cysts.

Demonstrated a genetic requirement for SRS9 in chronic persistence and reactivation control, moving it from marker to functional contributor.

Evidence ΔSRS9 knockout in a bioluminescent strain, oral and intraperitoneal mouse infection, bioluminescence imaging, brain cyst counts, and dexamethasone-induced reactivation

    Open questions at the time
    • Molecular mechanism by which SRS9 promotes persistence or restrains reactivation unknown
    • No host receptor or interaction partner identified
  5. 2018 High

Identified the upstream transcriptional switch enforcing SRS9 stage-specificity, explaining how tachyzoite-stage repression is achieved.

Evidence Genetic deletion of the AP2IV-4 locus with protein detection of ectopic SRS9 (and other bradyzoite proteins) in tachyzoites plus mouse infection

    PMID:29718996

    Open questions at the time
    • Whether AP2IV-4 binds the SRS9 promoter directly or acts indirectly not established
    • cis-regulatory elements of SRS9 not mapped
  6. 2019 Medium

Confirmed the SRS9 promoter as a bona fide bradyzoite-stage-specific expression driver, providing a tool leveraging its stage restriction.

    PMID:31572690

    Open questions at the time
    • Functional readout attributed to GRA17, not SRS9 protein; SRS9's own role not tested here
    • Promoter architecture and regulatory element boundaries not defined

  • No identified host or parasite interaction partner
  • Mechanism linking surface expression to persistence and reactivation control unknown

2002 SRS9 was identified as a novel developmentally regulated gene encoding a putative surface protein (SAG1-related protein) that is upregulated during tachyzoite-to-bradyzoite differentiation, as determined by microarray analysis of ~4,400 Toxoplasma cDNAs. Microarray expression profiling with relative transcript abundance measurement Eukaryotic cell Medium 12455982
2005 Constitutive expression of the normally bradyzoite-specific SRS9 in tachyzoites (SRS9c mutants) elicited a SRS9-specific immune response marked by IFN-γ production, which correlated with a continual decrease in the number of SRS9c cysts persisting in the brain, demonstrating that stage-specific expression of SRS antigens is a key mechanism for establishing and maintaining optimal parasite persistence. 2007 SRS9 protein is the dominant surface antigen recognized by the bradyzoite-specific P36 monoclonal antibody, confirmed by stable expression of SRS9 in BSR4-negative strains and immunoprecipitation experiments showing direct P36 mAb–SRS9 interaction. 2007 Deletion of SRS9 (ΔSRS9) in a bioluminescent Toxoplasma strain showed that during chronic infection, ΔSRS9 cyst loads substantially decreased compared to wild type, demonstrating a role for SRS9 in maintaining parasite persistence in the brain. Additionally, immunosuppressed chronically infected mice with ΔSRS9 showed intestinal reactivation within 8–9 days versus 2 weeks for wild type, indicating SRS9 plays a role in controlling reactivation in the intestine. 2018 The ApiAP2 transcription factor AP2IV-4 acts as a transcriptional repressor of SRS9 in tachyzoites; deletion of the AP2IV-4 locus resulted in aberrant expression of SRS9 (alongside other bradyzoite-specific proteins BPK1, MCP4, CST1) in replicating tachyzoites, demonstrating that AP2IV-4 normally suppresses SRS9 expression during acute infection. 2019 The SRS9 promoter was used to drive bradyzoite-specific expression of GRA17, and complementation with GRA17 under the SRS9 promoter drastically increased bradyzoite viability inside cysts, demonstrating that the SRS9 promoter is active specifically in the bradyzoite stage and can drive functional bradyzoite-stage gene expression.
