Synthesis pass · prose summary of the discoveries below

GRA9 is a Toxoplasma gondii dense granule protein that shapes the parasitophorous vacuole and cyst-wall environment and modulates host immunity during infection (PMID:15491588, PMID:31266861). After secretion from the anterior end of the parasite, GRA9 associates with the intravacuolar network (IVN) of tubular membranes, existing in both soluble and membrane-associated forms; its amphipathic and hydrophobic alpha-helices account for these dual solubility properties despite an overall hydrophilic character (PMID:15491588). During bradyzoite differentiation GRA9 dynamically redistributes to the cyst periphery and then into the inner layers of the maturing cyst wall, where it associates with cyst-wall components including the pseudokinase BPK1, and its loss slows the accumulation of cyst-wall proteins at the periphery (PMID:23291621, PMID:31619500). Consistent with this role, deletion of gra9 in type II strains causes severe chronic-stage cyst defects in vivo and impairs plaque formation, egress from the vacuole, and virulence, whereas the type I RH knockout instead shows an invasion defect, defining a strain-specific requirement (PMID:31266861, PMID:30849416). Beyond the parasite, the GRA9 C-terminal domain binds host NLRP3 through residue Q200 and blocks ASC recruitment to NLRP3, suppressing inflammasome assembly and promoting an anti-inflammatory M2 macrophage state (PMID:33182702).

2004 GRA9 is a dense granule protein (318 aa, 41 kDa) that, after secretion from the anterior end of the parasite, associates with the intravacuolar network (IVN) of tubular membranes within the parasitophorous vacuole. Unlike GRA2 or GRA6, GRA9 does not transit through the posterior invaginated pocket where the IVN first assembles. Within dense granules GRA9 exists in both soluble and insoluble states, but is secreted only as a soluble form; inside the vacuolar space two forms are detected: a soluble form and a membrane-associated form. The dual properties are attributed to amphipathic and hydrophobic alpha-helices despite the protein being predominantly hydrophilic. 2013 GRA9 physically associates with bradyzoite pseudokinase 1 (BPK1), a cyst wall component, as identified by immunoprecipitation from in vitro bradyzoite cultures. GRA9 was co-precipitated along with MAG1, MCP4, and GRA8. Immunoprecipitation (Co-IP) from bradyzoite cultures followed by protein identification Eukaryotic cell Medium 23291621
2019 Genetic deletion of gra9 (IVN membrane-associated GRA) in the type II Pru strain caused severe defects in chronic-stage cyst development in vivo without affecting parasite growth rate or in vitro differentiation into cysts, placing GRA9 as required for successful chronic infection. 2019 During cyst development in vitro, GRA9 dynamically redistributes: by day 1 post-differentiation GRA9 colocalizes with Dolichos biflorus agglutinin stain at the cyst periphery; by day 2 it establishes a continuous matrix pattern inside the cyst; and GRA9 localizes predominantly in the inner layers of the cyst wall. Deletion of gra9 reduces the rate of accumulation of cyst wall proteins at the cyst periphery relative to the cyst interior. 2019 Knockout of GRA9 in the type II PLK strain (PLKΔGRA9) inhibited plaque formation and parasite egress from the parasitophorous vacuole, and significantly reduced virulence in mice. These phenotypes were rescued by complementation with a synonymous PLK gra9 allele. In contrast, RHΔGRA9 (type I) showed no difference in plaque formation or egress but had a defect in host cell invasion, indicating strain-specific roles for GRA9. 2020 GRA9 interacts with NLRP3 (NLR family pyrin domain-containing 3) in host macrophages. The C-terminal domain of GRA9 (GRA9C) is essential for this interaction and disrupts NLRP3 inflammasome assembly by blocking the binding of ASC to NLRP3. Residue Q200 within GRA9C is specifically required for the NLRP3 interaction and blockade of ASC conjugation. Recombinant GRA9C exerted anti-inflammatory effects and promoted M1-to-M2 macrophage conversion in vitro and reduced sepsis pathology in vivo.
