Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

SOD in Toxoplasma gondii denotes iron-containing superoxide dismutase (FeSOD) activity that forms part of the parasite's antioxidant defense against reactive oxygen intermediates generated by the host cell (PMID:7452149, PMID:2760759). The single-copy TgSOD gene encodes a 201-residue, 22.6 kDa FeSOD whose iron cofactor was confirmed by atomic absorption mass spectrometry and EPR after recombinant expression in a SOD-deficient E. coli mutant, and the protein is present in both tachyzoite and bradyzoite stages, consistent with a role in intracellular growth of both developmental forms (PMID:10743616). Endogenous SOD activity, alongside catalase and glutathione peroxidase, is measurable in parasite lysates and is proposed to blunt macrophage oxygen-dependent killing by neutralizing superoxide and hydrogen peroxide (PMID:7452149, PMID:2760759). A second, distinct FeSOD, TgSODB2, is targeted to the mitochondrion via an unusual bipartite sequence: a hydrophobic N-terminal segment resembling a signal peptide is coupled to a downstream presequence, and only in this combination is ER translocation suppressed so that the protein is imported into the mitochondrion (PMID:14576360). Mutating the sole charged residue in the hydrophobic segment restores ER translocation and redirects the reporter to the apicoplast, while removal of the hydrophobic segment leaves the presequence fusion cytoplasmic (PMID:14576360).

Open questions

Synthesis pass · forward-looking unresolved questions
  • Whether TgSOD and TgSODB2 are individually required for resistance to host-derived reactive oxygen species and for parasite virulence remains unresolved.
  • No genetic knockout or complementation phenotype reported for either SOD
  • Non-redundancy between cytoplasmic and mitochondrial FeSODs untested

2000 The T. gondii SOD gene encodes a 201 amino acid, 22.6 kDa iron-containing superoxide dismutase (FeSOD), with a gene structure of four exons and three introns, occurring as a single-copy gene. Recombinant protein expressed in an E. coli double mutant lacking MnSOD and FeSOD confirmed iron as the metal cofactor by atomic absorption mass spectrometry and electron paramagnetic resonance. The protein was detected in both bradyzoite and tachyzoite forms, suggesting a role in intracellular growth of both developmental stages. 2003 TgSODB2 is a second, distinct mitochondrial iron-containing SOD in T. gondii with an unusual bipartite targeting sequence: a hydrophobic N-terminal segment (resembling a signal peptide) followed by a presequence. The hydrophobic segment alone can target a reporter to the secretory pathway, but when coupled to the presequence it is prevented from directing ER translocation and the protein is imported into the mitochondrion. Mutating the sole charged residue in the hydrophobic segment restores ER translocation and redirects the reporter to the apicoplast. Removal of the hydrophobic segment results in cytoplasmic localization of the presequence-reporter fusion. 1980 T. gondii tachyzoites contain endogenous superoxide dismutase (SOD), glutathione peroxidase, and catalase activities measurable in parasite lysates. The presence of these scavenger enzymes within the parasite is proposed to reduce the effectiveness of macrophage oxygen-dependent antimicrobial activity by neutralizing reactive oxygen intermediates (O2- and H2O2) generated by the host cell. Enzymatic activity assays in T. gondii lysates; aminotriazole inhibition experiments; comparison with acatalasemic mouse macrophages The Journal of experimental medicine Medium 7452149
1989 T. gondii tachyzoites are rich in SOD, catalase, and glutathione peroxidase as measured in cell-free systems. T. gondii is susceptible to H2O2 (50% growth inhibition at ~56 µM) and to products of xanthine-xanthine oxidase (OH· and singlet O2), but the parasite's endogenous SOD and other scavengers help protect it from O2-mediated killing. Cell-free enzymatic scavenger assays, H2O2 dose-response growth inhibition, xanthine-xanthine oxidase killing assay with specific quenchers/scavengers The Journal of parasitology Medium 2760759
