Synthesis pass · prose summary of the discoveries below

Toxoplasma gondii LDH2 is a bradyzoite-specific lactate dehydrogenase that supports the parasite's metabolic and developmental transition to the chronic cyst stage (PMID:9016946, PMID:29028143). It arose as a homolog of the constitutive tachyzoite enzyme LDH1 (71.1% identity), but its mRNA and cytoplasmic protein are restricted to the bradyzoite stage, with the LDH1-to-LDH2 isoenzyme switch occurring early during tachyzoite-to-bradyzoite conversion (PMID:9016946, PMID:12350375). As an enzyme, recombinant LDH2 displays broad substrate specificity, preferring 3-phenylpyruvate over pyruvate, efficiently using the NAD analog APAD, and — unlike LDH1 — lacking substrate inhibition; it is competitively inhibited by gossypol derivatives, dihydroxynaphthoic acids, and N-substituted oxamic acids (PMID:11704270). Functionally, LDH2 contributes to lactate fermentation that supplements energy production under physiological low-oxygen conditions, and its loss impairs chronic-stage cyst formation in vivo (PMID:29028143). Stage-specific expression is controlled at multiple levels: the LDH2 promoter is transcriptionally upregulated under bradyzoite-inducing alkaline conditions (PMID:9016946), its transcript abundance depends on the ATP-dependent chromatin-remodeling subunit TgRSC8 (PMID:21655329), and intron retention generates unspliced LDH2 transcripts during the stage transition (PMID:25777509).

1997 LDH2 gene encodes a lactate dehydrogenase homolog with 71.1% amino acid identity to LDH1; LDH2 mRNA is exclusively detected in the bradyzoite stage, while LDH1 mRNA is detected in both tachyzoites and bradyzoites. A reporter construct carrying the LDH2 promoter sequence was significantly upregulated under alkaline pH (pH 8.2, bradyzoite-inducing conditions), demonstrating that LDH2 expression is transcriptionally regulated in a stage-specific manner. Northern blot/RT-PCR for mRNA detection, isoelectric focusing and immunoblot for protein isoform analysis, chloramphenicol acetyltransferase (CAT) reporter gene assay with LDH2 promoter under alkaline pH conditions Gene High 9016946
2001 Recombinant LDH1 and LDH2 from T. gondii were characterized kinetically: both exhibit broader substrate specificity than Plasmodium falciparum LDH (pLDH); LDH2 utilizes 3-phenylpyruvate as a better substrate than pyruvate; both LDH1 and LDH2 efficiently use the NAD analog APAD; LDH2 (unlike LDH1) does not exhibit substrate inhibition despite having an identical M163 residue at the cofactor binding site. Both isoforms are competitively inhibited by gossypol and derivatives, dihydroxynaphthoic acids, and N-substituted oxamic acids. 2002 LDH2 protein is exclusively expressed in the cytoplasm of bradyzoites (not in tachyzoites or coccidian stages), as determined by immunocytochemistry and immunoelectron microscopy. The isoenzyme switch from LDH1 to LDH2 occurs at an early stage of tachyzoite-to-bradyzoite conversion when bradyzoites are actively proliferating. 2004 Knockdown of LDH2 (and LDH1) expression in stable transgenic parasites impaired tachyzoite and bradyzoite differentiation in vitro and severely reduced tissue cyst formation in a murine model. LDH expression is required for parasite differentiation and establishment of chronic infection. 2009 Over-expression of TgLDH2 (or TgLDH1) in tachyzoites did not affect enzymatic activity, growth, or virulence under standard conditions, but enhanced bradyzoite differentiation (increased cyst structure formation in vitro) under alkaline conditions, suggesting TgLDH2 has a physiological role in differentiation beyond glycolytic metabolism. 2015 Unspliced (intron-retained) forms of LDH2 transcripts are produced during tachyzoite-to-bradyzoite stage transition, suggesting an intron retention mechanism contributes to rapid exchange of LDH isoforms during developmental adaptation. 2017 Deletion of LDH2 alone impaired chronic-stage cyst burdens in vivo without affecting acute virulence or bradyzoite differentiation in vitro. Deletion of both LDH1 and LDH2 caused more severe defects in cyst formation. The double-knockout (Δldh1 Δldh2) replicated more slowly under physiological oxygen levels (3%) and was more susceptible to the oxidative phosphorylation inhibitor oligomycin A, indicating LDH2 contributes to lactate fermentation that supplements energy production when oxygen is limited. 2017 Deletion of LDH2 alone (Δldh2) impaired chronic-stage cyst burdens in vivo without affecting acute virulence or in vitro bradyzoite differentiation, whereas deletion of LDH1 reduced both acute virulence and bradyzoite differentiation. Combined deletion of LDH1 and LDH2 induced more severe defects in chronic cyst formation, but no growth defect was observed for any single or double mutant. 2011 TgRSC8, a subunit of an ATP-dependent chromatin remodeling complex (RSC/SWI-SNF ortholog), controls LDH2 transcript levels. In the C9 insertional mutant with reduced TgRSC8, steady-state LDH2 mRNA levels were significantly decreased; restoration of TgRSC8 expression by complementation significantly increased LDH2 transcript levels.
