Mechanistic narrative

Synthesis pass · prose summary of the discoveries below

GRA5 is a Toxoplasma gondii dense granule protein that is secreted into the parasitophorous vacuole and functions as a structural component of the vacuolar and cyst membranes across the parasite life cycle (PMID:8515776, PMID:8344403, PMID:10198072). It is initially secreted as a soluble form into the PV lumen and subsequently inserts into the parasitophorous vacuole membrane (PVM) as a transmembrane protein with its N-terminus facing the host cytoplasm and its C-terminus in the vacuole lumen; deletion within its single hydrophobic transmembrane domain abolishes PVM association without disrupting granule packaging or soluble secretion (PMID:10198072). Its N-terminal ectodomain is required both to prevent premature membrane integration inside the parasite and to direct correct post-secretory sorting and insertion into the vacuolar membrane, a soluble trafficking behavior that is parasite-specific, since in mammalian cells GRA5 inserts directly into the plasma membrane as a type I integral membrane protein (PMID:18631244). Within the parasite and PV soluble fraction GRA5 assembles into high-molecular-weight oligomeric complexes with GRA3, GRA6, and GRA7, retaining only an association with GRA7 after PVM insertion (PMID:17959262). During bradyzoite differentiation GRA5 transitions to the cyst membrane and cyst wall, where it is required for normal cyst wall maturation and, in the ME49 strain, for tissue cyst formation in vivo (PMID:8344403, PMID:31941814, PMID:37426671). GRA5 also contributes to immune evasion by suppressing MHC-I presentation of PV-localized antigens to CD8+ T cells (PMID:31555296), and its soluble N-terminal domain modulates dendritic cell migration via CCR7 (PMID:23033174). GRA5 is dispensable for the tachyzoite lytic cycle in vitro (PMID:11522359).

1993 GRA5 (P21) is encoded by an intron-less gene of 834 bp and the deduced protein contains a 25-amino-acid N-terminal signal peptide and a second hydrophobic domain consistent with a transmembrane region; ultrastructural studies confirmed association of GRA5 with the parasitophorous vacuole membrane (PVM). cDNA/genomic cloning, sequencing, peptide microsequencing of native antigen, ultrastructural immunolocalization Molecular and biochemical parasitology High 8515776
1993 In bradyzoites, GRA5 is specifically targeted to the delimiting membrane of the cyst wall, whereas GRA2 preferentially associates with the ground substance and tubular network, demonstrating selective sorting of dense granule contents after exocytosis. 1996 The GRA5 promoter contains repeated motifs (A/TGAGACG) within 156–30 bp upstream of the transcription start site that are critical for promoter activity, as demonstrated by deletion analysis and mutation of these elements; this cis-acting element is shared with GRA1, GRA2, and GRA6 promoters. 1997 In sporozoites, GRA5 is the only dense granule protein secreted during host cell invasion into the first parasitophorous vacuole (PV1), becoming associated with the PV1 membrane, whereas GRA1, GRA2, and GRA4 are retained in cytoplasmic granules and not exocytosed during PV1 formation.
