GRA17 is a Toxoplasma-secreted dense granule protein that localizes to the parasitophorous vacuole membrane (PVM) and mediates passive transport of small molecules across the PVM, functioning as a large non-selective pore. GRA17-deficient parasites exhibit aberrant PV morphology, reduced PVM permeability, structural instability, slow proliferation, and avirulence in mice. GRA17 functions synergistically with GRA23, and exogenous expression of GRA17 in Xenopus oocytes alters membrane conductance consistent with a large non-selective pore. |

GRA17 is functionally related to the Plasmodium EXP2 translocon protein and is conserved across PV-residing Apicomplexa. Complementation of GRA17-deficient Toxoplasma parasites with Plasmodium EXP2 rescues the GRA17-deficient phenotypes (PV morphology, permeability, growth), establishing functional conservation between these pore-forming proteins. |

GRA17 plays an important role in bradyzoite viability inside cysts. Deletion of GRA17 in the type II ME49 strain produced enlarged 'bubble vacuoles' with reduced PVM permeability. ME49Δgra17 parasites formed cysts in vitro at comparable rates to wild-type, but bradyzoite viability inside cysts was significantly reduced. Complementation with GRA17 expressed from a bradyzoite-specific promoter (SRS9) drastically increased bradyzoite viability, demonstrating GRA17's importance in the cyst stage. |

Deletion of gra17 in Toxoplasma gondii type II Pru strain abrogated acute infection and blocked cyst formation, demonstrating GRA17's role in establishing chronic infection. Pru:Δgra17 mutants developed PVs with abnormal morphology and exhibited a reduced growth rate compared to wild-type. |

In vivo CRISPR screen results suggest a role of GRA72 for normal GRA17/GRA23 localization, indicating that GRA72 is required upstream of GRA17 for its proper targeting to the PVM. |

GRA47 interacts with GRA72 (identified by immunoprecipitation), and both GRA47 and GRA72 are pore-forming proteins that regulate PVM permeability alongside GRA17. Overexpression of GRA17 (or GRA23 or GRA72) partially rescued aberrant PV morphology in Δgra47 parasites, and P. falciparum PfExp2 (an ortholog of GRA17/GRA23) fully rescued PV morphological defects in Δgra47, Δgra72, and Δgra17 strains, indicating partial functional redundancy among this family of pore-forming GRA proteins. |

Co-immunoprecipitation demonstrated interaction of GRA47 with GRA72. Loss of GRA72 affected PV morphology, parasite growth, and infectivity. Overexpressing GRA17 partially rescued PVs with aberrant morphology in Δgra47 and Δgra72 parasites, while PfExp2 fully rescued the PV morphological defect in Δgra17, Δgra47, and Δgra72 strains. |

GRA1 inactivation impaired the secretion of GRA17 to the PVM and disrupted the intravacuolar network (IVN), which mediates transport of molecules in and out of parasites. Mislocalization of GRA17 (and other GRA proteins) caused by GRA1 depletion resulted in a strong defect in absorbing nutrients (glucose, pantothenate) from host cells, placing GRA1 upstream of GRA17 in the nutrient uptake pathway. |

AAP7 (apical annuli protein 7) is required for efficient secretion of GRA17 from dense granules. AAP7 depletion causes reduced GRA17 secretion, leading to 'bubble' vacuoles characteristic of GRA17 loss. Overexpression of GRA17 overcomes AAP7 depletion and reduces bubble vacuoles, identifying reduced GRA17 secretion as the critical defect downstream of AAP7 loss. |
